Optimisation des conditions hydrobiologiques pour la conservation de l'huître creuse Crassostrea gigas en système re-circulé ArchiMer
Buzin, Florence.
The maintenance of bivalve commercialisation du ring periods when sales are prohibited represents a major economic issue for shellfish-farming. To ensure their survival and to preserve the oyster Crassostrea gigas quality, their storage in land-based systems has been studied i) out of water in a cold room at 3°C with controlled humidity, enabling the bivalve sensory quality to be retained for 15 days, and ii) in recirculating systems in immersed conditions for 30 days. For the latter, the oysters were stored at 167 kg.m3 density in a tank equipped with an air-lift system, with water treated with UV and thermoregulated. A minimum of food was supplied to avoid mass tissue loss. A temperature of 16°C was defined as optimal to limit mortality and to avoid...

Oyster hemocytes express a proline-rich peptide displaying synergistic antimicrobial activity with a defensin ArchiMer
Gueguen, Yannick; Romestand, Bernard; Fievet, Julie; Schmitt, Paulina; Destoumieux Garzon, Delphine; Franck, Vandenbulcke; Philippe, Bulet; Bachere, Evelyne.
A cDNA sequence that encodes a 61-amino acid polypeptide precursor with homologies to proline-rich antimicrobial peptides (AMPs) was identified in the oyster Crassostrea gigas. After release of a hydrophobic signal peptide. the resulting 37-amino acid peptide, Cg-Prp, is composed of an acidic region and a cationic proline-rich region. To evaluate the biological properties of Cg-Prp, multiple proline-rich peptides corresponding to putative processing of the full-length Cg-Prp were synthesized. A limited antimicrobial activity was observed for two of them, which also showed strong synergistic antimicrobial activity with Cg-Def, a defensin from C gigas. To our knowledge, this is the first evidence of synergy between a defensin and another AMP in an...

Pacific oyster (Crassostrea gigas) hemocyte are not affected by a mixture of pesticides in short-term in vitro assays ArchiMer
Moreau, Pierrick; Burgeot, Thierry; Renault, Tristan.
Pesticides are frequently detected in estuaries among the pollutants found in estuarine and coastal areas and may have major ecological consequences. They could endanger organism growth, reproduction or survival. In the context of high mortality outbreaks affecting Pacific oysters, Crassostrea gigas, in France since 2008, it appears of importance to determine the putative effects of pesticides on oyster susceptibility to infectious agents. Massive mortality outbreaks reported in this species, mainly in spring and summer, may suggest an important role played by the seasonal use of pesticides and freshwater input in estuarine areas where oyster farms are frequently located. To understand the impact of some pesticides detected in French waters, their effects...

Pollutant effects on Pacific oyster, Crassostrea gigas (Thunberg), hemocytes: Screening of 23 molecules using flow cytometry ArchiMer
Gagnaire, Beatrice; Thomas Guyon, H; Burgeot, Thierry; Renault, Tristan.
The shellfish industry is an important economic activity in France, occurring mostly in estuarine zones subject to pollution due to anthropogenic activities. The harmful effects of pollutants on species inhabiting these estuarine zones are not well known. Among marine species, bivalve mollusks---particularly Pacific oyster, Crassostrea gigas-may serve a model of interest. The species is sedentary and filter-feeding, which favors bioaccumulation of pollutants in their tissues. Oysters may be suitable for studies on disturbance by pollutants of physiological activities, among which defense mechanisms are poorly documented in bivalves. In this study, effects of pollutants on hemocyte functions were monitored in Pacific oyster, C. gigas. Hemocytes were exposed...

Summer mortality has been reported in the Pacific oyster, Crassostrea gigas, for many years in Japan, US and Europe. It affects both adults and juveniles and the causes of this phenomenon are complex. Environmental, physiological or pathological putative causes were studied all over the world. Recently, the MOREST project ((MORtalitEStivale) a multidisciplinary project (2001-2005) focused on his problem led to a better understanding of this phenomenon in France. The results showed in these geographical conditions, the main interactions between the oysters, their environment and certain pathogens.

QTL for resistance to summer mortality and OsHV-1 load in the Pacific oyster (Crassostrea gigas) ArchiMer
Sauvage, Christopher; Boudry, Pierre; De Koning, D. -j.; Haley, C. S.; Heurtebise, Serge; Lapegue, Sylvie.
P>Summer mortality is a phenomenon severely affecting the aquaculture production of the Pacific oyster (Crassostrea gigas). Although its causal factors are complex, resistance to mortality has been described as a highly heritable trait, and several pathogens including the virus Ostreid Herpes virus type 1 (OsHV-1) have been associated with this phenomenon. A QTL analysis for survival of summer mortality and OsHV-1 load, estimated using real-time PCR, was performed using five F-2 full-sib families resulting from a divergent selection experiment for resistance to summer mortality. A consensus linkage map was built using 29 SNPs and 51 microsatellite markers. Five significant QTL were identified and assigned to linkage groups V, VI, VII and IX. Analysis of...

Petton, Bruno.

Rapid accumulation of an interleukin 17 homolog transcript in Crassostrea gigas hemocytes following bacterial exposure ArchiMer
Roberts, S; Gueguen, Yannick; De Lorgeril, Julien; Goetz, F.
Interleukin-17 (IL-17) gene models have been found in the sequenced genomes of Strongylocentrotus purpuratus and Caenorhabditis elegans. However, there have been no published reports on the empirical cloning and characterization of any interleukin cDNAs in invertebrates. From a Pacific oyster (Crossostrea gigas) hemocyte cDNA library, two clones were obtained that encoded a protein similar to vertebrate IL-17s. The putative oyster IL-17 homolog (CgIL-17) was 27% identical to rainbow trout IL-17D, 21% to human IL-17D and 24% to an IL-17D-like gene model obtained from the annotation of the sea urchin genome. IL-17s from the oyster, sea urchin, trout and human, contained conserved cysteine residues found in all forms of IL-17 in mammals. Injection of bacteria...

Relationships between oyster mortality patterns and environmental data from monitoring databases along the coasts of France ArchiMer
Soletchnik, Patrick; Ropert, Michel; Mazurie, Joseph; Fleury, Pierre-gildas; Le Coz, Florence.
Oyster mortality was monitored at multiple sites along the French coastline (around 5 500 km) between 1993 and 2005. Mortality data for one- and two-year-old oysters were collected over 10–12 years in 39 oyster culture sites bordering 3 different "seas": the English Channel, Bay of Biscaye and Mediterranean. Combining these data with records from environmental monitoring databases, 11 of these sites had consistent chronological data sets including both environmental data and mortality records. Mortality in one-year-old oysters was clearly summer mortality (49% of their annual mortality) whereas mortality of two-year-olds occurred mostly in spring (51%). Analysis of variance revealed that "coastal area" was the main influence on mortality of one-year-olds...

RESCO - Réseau d'observations Conchylicoles : Campagne 2013 ArchiMer
Fleury, Elodie.
The French network RESCO was developed in 2009, following the widespread mortalities in French oysters stocks of Crassostrea gigas that killed a majority of young cultivated oysters in summer 2008. The main objective of this network was to develop a tool of estimation of the mortalities at national scale, through a standardized monitoring of sentinel oysters In 2013, similar batches of spat and 18 month old oysters from different origins were deployed in thirteen sites located all along the French coasts to measure the spatial and temporal evolution of the mortality and growth rates. At the same time, the program provides a high-frequency recording of environmental parameters and the detection of the main infectious agents associated to the oyster...

Scanning the genome of the Pacific cupped oyster, Crassostrea gigas, for adaptative differentiation during geographic range expansion ArchiMer
Rohfritsch, Audrey; Bierne, Nicolas; Boudry, Pierre; Heurtebise, Serge; Lapegue, Sylvie.
Originating from the north eastern Asia, Crassostrea gigas has been introduced and translocated, mainly for aquaculture purpose, into several European countries (from Norway to Portugal and in the Mediterranean Sea) (1). Although highly variable, the invasiveness pattern of C. gigas has been demonstrated in several countries and therefore considered as a pest or a noxious species in those areas (2 and 3).Our project aims at identifying the characteristics of such a flourishing species: can its success be explained by chance and/or global warming only or does it exhibit a more important potential of adaptation than other species? Therefore we developed a population genomics approach, known as "genome scan". It corresponds to the study of numerous loci...

Selective breeding to improve resistance against summer mortality in the Pacific oyster Crassostrea gigas : results after 3 generations ArchiMer
Boudry, Pierre; Degremont, Lionel; Bedier, Edouard; Samain, Jean-francois.
Summer mortality of adults and juveniles has been reported in the Pacific oyster, Crassostrea gigas, for many years and in several countries. The French multidisciplinary program "Morest" aims to investigate the causes of the summer mortality in Crassostrea gigas. Within this program, we designed multi-site field experiments to assess to what extent genetic variability exists for summer mortality in French populations of C. gigas and to determine whether selective breeding could improve survival.

Setting of a procedure for experimental fertilisation of Pacific oyster (Crassostrea gigas) oocytes ArchiMer
Song, Yun-peng; Suquet, Marc; Queau, Isabelle; Lebrun, Luc.
The quality of oyster spermatozoa has become an issue for modern aquaculture. However, reliable protocols used to assess gamete quality are lacking. The aim of this work is to define a standardised experimental fertilisation protocol for Pacific oyster oocytes. Six experiments have been carried out in this study. The optimal conditions for Pacific oyster experimental fertilisation have been defined: (1) oocytes can be conserved in seawater for at least 4 h before fertilisation. (2) oocyte concentration: 100-1000 ml(-1) seawater, (3) fertilisation volume: 10-100 nil, (4) spermatozoa: oocyte ratio: 400 to maintain discriminating conditions, (5) gamete contact time: longer than 10 mill and (6) fertilisation and incubation temperatures: both 19 degrees C. Setting of a procedure for experimental incubation of Pacific oyster (Crassostrea gigas) embryos ArchiMer
Suquet, Marc; Amourda, Christopher; Mingant, Christian; Queau, Isabelle; Lebrun, Luc; Brizard, Raphael.
Standardized experimental protocols designed to study Pacific oyster (Crassostrea gigas) gamete quality have not been previously published. Gamete sampling variations and confounding effect of interacting factors result in large variations between replicates, decreasing the effects of the studied factors. This work aims at defining a standardized procedure for incubation of oyster embryos designed for experimental purposes. In a first phase, four experiments were developed to improve embryo sampling and handling. They showed that a minimum of 50 embryos must be counted to decrease the variation between counting replicates. For reliable results, sampling must be carried out from 0 to 7 min after a careful agitation of seawater containing embryos...

Sources of paralytic shellfish toxin accumulation variability in the Pacific oyster Crassostrea gigas ArchiMer
Pousse, Emilien; Flye-sainte-marie, Jonathan; Alunno-bruscia, Marianne; Hegaret, Helene; Jean, Fred.
This study was designed to assess the contribution of feeding behavior to inter-individual variability of paralytic shellfish toxin (PST) accumulation in the Pacific oyster Crassostrea gigas. For this purpose 42 oysters were exposed for 2 days to non-toxic algae and then for 2 other days to the PST producer Alexandrium minutum. Individual clearance rate (CR) of oysters was continuously monitored over the 4 days using an ecophysiological measurement system. Comparison of CR values when exposed to toxic and non toxic algae allowed to estimate a clearance rate inhibition index (CRII). Toxin concentration of oysters was quantified at the end of the experiment. These data allowed to estimate the toxin accumulation efficiency (TAE) as the ratio of toxin...
